E i famosi vaccini? Roba superata!

Oramai, come ogni anno, l’influenza ci attende per regalarci dei momenti di relax! Facendoci rimanere a casa e impedendoci di stare all’aria aperta o in ambienti lavorativi, visto i sintomi che indeboliscono il nostro organismo e ci rendono contagiosi.

Tantissimi di noi per prevenire questi malanni utilizzano il Vaccino. Questi prodotti, che in origine erano progettati per combattere le malattie infettive, sono ottenuti da microorganismi e ottimizzati in laboratorio per veicolare quello che ci darà l’immunità dalla malattia.

EFFETTI COLLATERALI E SENSIBILITA’ INDIVIDUALE

È accertato che in alcuni individui, a cui viene somministrato il vaccino, possono svilupparsi delle reazioni collaterali sia locali, come l’arrossamento della zona dove è avvenuta l’iniezione, che generalizzati con febbre e malessere, perché il loro sistema immunitario ha reagito al farmaco. Queste possibili reazioni vengono studiate già nelle fasi di sviluppo della formulazione.

Reazioni più gravi sono imprevedibili e fortunatamente avvengono solo raramente. Gli effetti collaterali gravi possono essere individuati solo quando il vaccino viene somministrato a milioni di persone. In questo caso, entrano in gioco gli opportuni enti che si occupano della farmacovigilanza, i quali analizzeranno gli eventi avversi e prenderanno gli opportuni provvedimenti, che possono portare anche al ritiro del farmaco dal mercato.

CHE FARE? ECCO ANCHE IN QUESTO CASO VENGONO IN AIUTO LE NANOTECNOLOGIE

Di cosa stiamo parlando? Del vaccino universale basato su NANOPARTICELLE proteiche, sviluppato da una start up nata solo due anni fa in Gran Bretagna, che è stato testato sulle cavie di laboratorio e ha dato dei risultati migliori rispetto al vaccino stagionale, il quale deve essere ripetuto ciclicamente, in quanto i virus mutano, rendendo l’effetto di protezione temporaneo.

I vaccini stagionali hanno come target una glicoproteina presente sulla superficie del virus, l’emoagglutinina (HA), composta da una porzione esterna al virus chiamata “testa” e una interna detta “gambo”. La “testa” dell’HA è anche il punto cruciale della limitatezza del vaccino stagionale, in quanto questa proteina può variare ed è differente nei vari tipi di virus. L’idea del vaccino con nanoparticelle è quello di colpire la porzione di HA interna al virus, la quale si conserva nei virus e quindi permette di ottenere una protezione UNIVERSALE.

È da circa 50 anni che si sta cercando di ottenere un vaccino di questo tipo e forse siamo vicini ad una soluzione!

L’idea è di colpire il “gambo” della proteina della HA, che è condiviso dai virus influenzali, ma che non risulta stabile se iniettato così com’è. La soluzione dei ricercatori è stata di assemblare il “gambo” della HA in una nanoparticella proteica che funge da involucro a questo antigene. Una volta iniettate, le nanoparticelle vengono ingerite dalle cellule che hanno il compito di attaccare i corpi estranei, e da questo punto che poi si svolgerà una serie di reazioni, che porteranno allo sviluppo dell’immunità verso il patogeno.

Questo vaccino ha dimostrato una buona efficacia soprattutto contro l’influenza A (aviaria,suina), più infettiva nei nostri confronti (ovvero quella che colpisce più specie, a differenza dell’influenza di tipo B che è esclusiva dell’uomo).

I primi test sull’uomo sono stati eseguiti da un gruppo di ricerca dell’Università di Oxford, in cui si è somministrato il farmaco a base di nanoparticelle contro l’influenza A. I dettagli sono stati pubblicati sulla prestigiosa rivista scientifica Nature Communications.

SARA’ REALMENTE COMPATIBILE CON TUTTI?

Sembra che non ci sia stata nessuna incompatibilità o controindicazione del prodotto, ma ottenuti questi promettenti risultati, ora è necessario focalizzarsi sullo studio e l’approfondimento della sua sicurezza, oltre che alla sua funzionalità. Anche se non è stato specificato se individui che hanno particolari patologie siano esenti da effetti collaterali, bisogna valutare la TOLLERABILITA’ della sostanza iniettata in determinate quantità, e la questione è abbastanza soggettiva, potendo variare nei diversi contesti. Considerato che comunque il vaccino si propone come UNIVERSALE diamo fiducia a quanto detto finché non ci sarà una ufficializzazione su scala mondiale.

Infine, è necessario, se non essenziale per i ricercatori sviluppare delle tecniche che siano economiche o almeno equivalenti all’alternativa tradizionale, per la produzione di questo vaccino. Quando vi sarà la certezza della sicurezza e della produzione del vaccino, quest’ultimo potrà essere sottoposto all’analisi ed eventuale approvazione degli enti regolatori del farmaco come l’EMA (European Medicines Agency) e FDA (Food and Drug Administration) che ne consentiranno la produzione da parte delle case farmaceutiche e la sua dispensazione nei vari stati.

And the famous vaccines? Old stuff! Every year, the flu awaits to give us moments of "relaxation"! Making us stay at home and preventing us from going outdoors or at work, given the symptoms that weaken our body and make us contagious. For prevention of this disease, many of us use the Vaccine. These products, which were originally designed to fight infectious diseases, are obtained from microorganisms and optimized in laboratory to convey what will give us immunity from the flu.

SIDE EFFECTS AND INDIVIDUAL SENSITIVITY

It is established that in some individuals, to whom the vaccine is administered, local side reactions may take place, such as the redness of the area where the injection occurred, or generalized effects with fever and fatigue. This happens because their immune system reacted to the drug. These possible reactions are already studied in the development stages of the formulation. More serious reactions are unpredictable and fortunately they occur only rarely. Serious side effects can only be detected when the vaccine is given to millions of people. In this case, the appropriate agencies involved in pharmacovigilance come into play, analyzing the adverse events and take appropriate measures, which can also lead to the removal of the drug from the market.

WHAT CAN WE DO? WE CAN USE NANOTECHNOLOGIES TO HELP US!

What are we talking about? We talk about the universal vaccine based on PROTEIN NANOPARTICLES developed by a start up born only two years ago in Great Britain, which was tested on laboratory animals and gave better results than the seasonal vaccine, which must be repeated cyclically, because viruses mutate, so the protection from the vaccine effect is temporary. Seasonal vaccines target a glycoprotein present on the surface of the virus, called hemagglutinin (HA), which is composed from an outside portion (the "head") and an internal part (called "stem"). The "head" of HA is what creates the limitation of the seasonal vaccine, as this protein can mutate and you can find it in different form in various types of viruses. The fondamental idea of the nanoparticle vaccine is to hit the portion of HA inside the virus, which is preserved in the majority of viruses and therefore allows to obtain a UNIVERSAL protection. It is about 50 years that we are trying to get this type of vaccine and maybe we are close to a solution! The idea is to hit the "stem" of HA protein, which is shared by influenza viruses, but which is not stable if injected as it is. The researchers' solution was to assemble the "stem" of HA in a protein nanoparticle that acts as an envelope to this antigen. Once injected, the nanoparticles are ingested by the cells that have the task of attacking the foreign bodies, and from this point a series of reactions will take place, which will lead to the development of immunity to the pathogen. This vaccine has shown a good effectiveness especially against influenza A (avian, swine), more infectious towards us and that is also the one that affects most species, unlike the type B influenza which is exclusive to human. The first human tests were performed by a research group at the University of Oxford, where the drug based on nanoparticles was used against influenza A. The details were published in the prestigious scientific journal “Nature Communications”.

WILL IT BE REALLY COMPATIBLE WITH ALL?

It seems that there has been no incompatibility or contraindication of the product, but obtained these promising results, now it is necessary to focus on the study and the deepening of its safety, as well as its functionality. Although it has not been specified whether individuals with particular pathologies are free from side effects, the TOLERABILITY of the injected substance must be evaluated in certain quantities, and the issue is quite subjective, because can vary in different contexts. Considering that in any case the vaccine is proposed as UNIVERSAL, we trust what has been said until there is an officialization on a world scale. Finally, it is necessary, if not essential for researchers to develop techniques for the production of this vaccine, that can be cheaper or at least equivalent to the traditional alternative. When there will be the certainty of safety and production of the vaccine, it may be subjected to the analysis and possible approval of the regulatory agencies, such as the EMA (European Medicines Agency) and FDA (Food and Drug Administration) that will allow production by pharmaceutical companies and its dispensation in the various states.
