This photo looks like a bunch of balloons, but it’s not. It’s a brain! As a part of one of my neuroscience lessons, I was asked to present a research about anything that involves brain cells. I did non know which research to present, but I was sure that I will show this pictures to my class. Like you can imagine, my lecturer was full of criticism. He asked me a lot of niggling questions like “why should you use this complicated staining?" But you obviously know, I was happy. Because I showed everyone this beautiful pictures. Camillo Golgi developed the first brain staining technique at 1873 (he deserves a whole post just about his ironic science story). Golgi called his staining “black reaction”, because the neurons were stained at black - not colorful like this photo, but also very neat. It’s been more than a hundred years until this super-cool inventing of the “Brainbow”. In this method, we genetic engineer mice. First of all, the mice need to be cre/lox - able to manipulate DNA by cutting or inversing. We also need the mice to have the staining - fluorescent proteins (XFPs). The sophisticated method uses the fact that every neuron get a few XFPs, when each XFP is a construct of red, yellow and blue. Between the colors there are lox sites. If the XFP is cut at the first site, the yellow color will be expressed. At the second site the blue color will be expressed, and if the XFP will stay uncut (Did you follow?) the red color will be expressed because it is the first one. Every neuron change the XFPs in a different way, thereby gets a different number of XFPs for each color. For example, a neuron that gets a lot of red XFPs, a few yellow XFPs, and no blue XFPs at all - will be stained in orange. Compared to a neuron that gets a few red XFPs, and a lot of yellow and blue XFPs - this neuron will be stained in green. Brainbow is so brilliant because every neuron can get a random color of more then a hundred colors! So how we can use Brainbow? If we ignore our wish to hang some of this photos at the livingroom… One of the interesting topics in brain science is the pathways of the neurons. Which neurons are connected (have synapses) together? Where are the strong connects? Which activity (trauma, memory, love) affects the connecting between the neurons? and so on. Like you can see if you scroll a little, Brainbow literally shows the synapses and the pathways of neurons. Look at the firs picture, that is divided to five pictures - form A to E. Can you recognize which brain region is at every part? After you had a few thoughts, here are the answers: A - We see neurons with wide trees (this trees are called dendrites) that are contacting a lot of other cells. This are Purkinje cells at the cerebellum. B - The neuron are very density and also very long (this long part is calls axon). This is the hippocampus. C - You can’t miss the big cells that are quit spacious from each other - like you probably thought, this is the brain stem. D- The neuron are organized in layers, from top to down. This is the outer part of the brain - the cortex. E - Looks a a lot like B with the long axons and the destiny, it is indeed a different part of the hippocampus. Maybe you have not been able to identify, because you are usual to think of the human brain. This pictures are taken from mice brain, that is different. We can also see the connection of the overall brain, like at the ending photo. I think you already saw that I have two big passions: brain and art. So fun you can combine them! תסתכלו על החץ הלבן. רואים שיש משהו ירוק שמחבק את המשהו האדום? לא תאמינו מה זה. במסגרת הלימודים שלי, התבקשתי להכין פרזנטציה על נושא מסויים שקשור לתאי עצב. לא ידעתי על איזה נושא לדבר, אבל בדבר אחד הייתי בטוחה - את התמונות האלה אני מראה. כמו שאתם מתארים לעצמכם, המרצה היה מלא בביקורת, ושאל כל מיני שאלות קטנוניות כמו "למה להשתמש בצביעה המסובכת הזו?", אבל אני, אני הייתי מאושרת. כי הראיתי לכולם את התמונות היפות האלה. את הצביעה הראשונה של המוח גילה אדם בשם קמילו גולג'י בשנת 1873 (הוא עוד יקבל פוסט נפרד בגלל האירוניה שקשורה אליו). גולג'י קרא לצביעה שלו "תגובה שחורה", פשוט כי הנוירונים נצבעו בצבע שחור. עברו יותר מ100 שנה עד להמצאה הסופר-מגניבה ופוטוגנית של הBrainbow. בשיטה זו, מהנדסים עכברים גנטית. קודם כל העכברים צריכים להיות cre/lox, כלומר עכברים שיכולים להפריד את רצף הDNA ולבצע עליו מניפולציות כמו חיתוך או היפוך. אנחנו גם צריכים שיהיה לעכברים את החלבון שצובע את המוח - XFPs. השיטה המתוחכמת גורמת לכך שכל נוירון יקבל מספר חלבונים פלורוסנטיים צובעים, כאשר כל חלבון כזה הוא רצף של צבעים שבנוי מאדום, צהוב וכחול. בין הצבעים נמצאים אתרי חיתוך. אם נחתוך את החלבון הפלורוסנטי בחיתוך הראשון, יתבטא הצבע הצהוב. בחיתוך השני יתבטא הצבע הכחול, ואם לא נחתוך בכלל (עקבתם?) האדום יתבטא כי הוא הראשון. אקראית, כל נוירון חותך באופן שונה את החלבונים הפלורוסנטיים, ובעצם מקבל מספר חלבונים שמתבטאים בכל צבע. לדוגמא, נוריון שיקבל כמות גדולה של אדום, כמות קטנה של צהוב, ובכלל לא כחול - יצבע בכתום. לעומתו, נוירון שיקבל מעט מאוד אדום, הרבה צהוב, והרכבה כחול - יצבע בירוק. הגאונות של השיטה היא בכך שכל נוירון מקבל צבע אקראי (אפשר ליצור כמאה צבעים כאלה). למה זה טוב? אם לרגע נתעלם מהרצון שלנו לתלות כמה מהתמונות האלה בסלון... אחד הדברים שמעניינים ביותר בחקר המוח, הוא הנתיב של הנוירונים. כלומר, איזה נוירונים נקשרים (יוצרים סינפסות) ביחד, איפה הקשרים הם חזקים, איך פעילות (טראומה, זיכרון, אהבה) משפיעה על הקשרים בין הנוירונים, וכן הלאה. הצביעה הצבעונית הזו מראה לנו בצורה מאוד חיה את הנתיבים של הנוירונים ואת הסינפסות ביניהם. לדוגמא, אם תחזרו לתמונה ששמתי למעלה (שמחולקת לחמש תמונות מA עד E) האם תוכלו לזהות איזה חלק מוחי נמצא בכל חלק? אחרי שחשבתם קצת, הנה התשובות: בתמונה A אפשר לראות תאים עם עץ רחב (לעץ הזה קוראים דנדריט), שמתקשרים עם הרבה תאים אחרים. אלו נוירונים בצרבלום שנקראים תאי פורקנייה. בתמונה B אפשר לראות שהנוירונים מאוד צמודים אחד לשני והם מאוד ארוכים (החלק המוארך הזה נקרא אקסון), זהו ההיפוקמפוס. בתמונה C, התאים מאוד גדולים ונמצאים ברווח אחד מהשני - בטח זיהיתם שזה גזע המוח. בתמונה D הנוירונים מסודרים בשכבות מלמעלה למטה, זהו החלק החיצוני של המוח - הקורטקס. תמונה E מזכירה מאוד את תמונה B, עם הנוירונים הצמודים והאקסונים הארוכים. אכן, זהו חלק אחר של היפוקמפוס. אולי לא הצלחתם לזהות, כי אתם רגילים לחשוב על המוח האנושי. התמונות האלו לקוחות ממוח של עכברים. אם נסתכל על כל המוח מלמעלה, נוכל גם לראות את הקשר בין החלקים השונים, כמו בתמונה הזו: ![](https://steemitimages.com/DQmTtkcX6uRtzot7KUhwpvYeUaV3tJSsuXHMf3khL15dnA1/image.png) בטח כבר הבנתם עליי שיש לי שתי אהבות גדולות בחיים: מוח ואמנות. איזה כיף שאפשר לשלב ביניהן! ספרו לי גם מה מעניין אתכם, הכל משתלב עם המוח :)
